EXPERIMENT 2 : ENZYME ASSAY
1. Explain the importance of standard reference?
The importance of standard reference are it act as reference to determine the concentration of a sample especially when we are constructing the Standard Curve. By having this standard reference, we can compared the results of samples. For this experiment, we are measured the concentration of starch.
2. What is function of Iodine solution ?
Iodine solution useful to determine the presence of starch in a solution. If the solution contain starch, the colour of solution will changed from yellow color turn to blue black.
3.Explain why starch turn blue when react with iodine?
Starch are composed of amylose and amylopectin. For amylose, there are specific linkages which can results in the formation of coiled linear region. Therefore, when the iodine ion fits into the core , it will stabilizing the shape by making contact with the sugar monomers and this will produce the colour changes into blue solution.
4. Provide the relationship between substrate concentration with enzyme activity?
The rate of enzyme activity or reaction is influenced by substrate concentration. The greater value of substrate concentration, the more higher the enzyme activity, Based on this experiment, the increase of reaction will cause Vmax value is approached. However, the when the concentration of substrate is reaching the Vmax point, the enzyme will stop binding with the substrate because there are no production of enzyme. Therefore, the rate of reaction will be constant.
5.What is Km?
Km defined as the substance concentration at 1/2 of the maximum velocity (Vmax).
6.What is Vmax?
Maximum velocity (Vmax) defined as the rate of the enzyme catalyzed a reaction. Normally it happens when all enzyme active sites are saturated with substrates.
7. Conclude the effect of temperature on product.
Based on this experiment, the higher velocity of rate of reaction for enzyme is 28 degree Celsius. The optimum temperature for enzyme is 37 degree Celsius which is same as our body temperature. Therefore, the tendency for the enzyme to denatured are higher if the temperature increase. However, on this experiment, the enzyme still not denatured because it only approaching 28 degree Celsius.
8.Learning experience.
For me, I am more concern about the group performance. How are we managed our group discussion and how are we dividing our works. Each of the member should give the best commitment for the group and always help each other. In a nutshell, what ever it is, I always thank to my fellow members because we are able to be as a group. THEY ARE AWESOME !
